Windows defrag utility Disk Defragmenter does not recognize mobile phone’s micro SD card, so you will need a third-party utility to do the job.

How to defrag your micro or mini SD card?
Plug-in you mobile phone micro/mini SD card on your computer. You will require SD card reader for this. If you have a smart phone, just plug-in the mobile phone using data cable and make sure the setting in you phone is to make it detected as storage device instead of mobile phone.
Click on Add button to select the SD card or individual files in the SD card.
Click on Analyze and later on Defragment button to defragment the SD card.
